Analysis of L643 SNP

SUMMARY OF SNP (AUGUST, 2012)

Classification of SNP: Near Private.

Estimated Breadth of SNP (6 of 9 match - over 70 % match): Only nine submissions. All three positive submissions have been with 8 of 9 matches or higher fingerprint matches. There are five excellent testing candidates with the surname of Matheson and McKenzie dominating the submissions that match the L643 fingerprint. Another testing candidate, a Robertson submission, is the best testing candidate for determining the boundary conditions of the L643 SNP. All untested candidates belong to the McKenzie cluster labeled "Group 14 - Colonial Maryland" found in the McKenzie Surname Project and all Matheson submissions belong to "Group Shiness" found in the Matheson Surname Project.

Estimated Breadth of SNP (speculative estimate of positive submissions) = 15 = 7 x 98 % (9 of 9) + 1 x 95 % (8 of 9) + 1 x 92 % (7 of 9) + 12 x 20 % (6 of 9) + 77 x 5 % (5 of 9). These optimisticly high percentages at low fingerprint matches are based of the rare marker value of 442 <= 11 and 481 <= 21 appearing in lower matches which could radically expand the scope of the L643 fingerprint.

439 <= 13 and 454 >= 12 are post L643 mutations that track the McKenzie submissons and 390 >= 25, 391 <= 10 and 448 <= 18 are post L643 submissions that track the Matheson submissions. The Orem submission tracks the McKenzie key marker values. This means that the Orem submission is either pre-McKenzie surname creation or post-McKenzie surname creation which would make the Orem branch become a NPE sub-branch of the McKenzie branch. The Robertson matches only one of the McKenzie marker values which implies a pre-McKenzie surname creation branch above the McKenzie branch or a post McKenzie creation sub-branch with a backwards mutation from 439 < = 13 to the original L21 MRCA.

Scope of Testing within Fingerprint: Average - Three out of the nine known 67 marker submissions have been tested and all have tested positive. There are also ten additional 37 marker Matheson submissions that have high fingerprint matches with markers within the L643 fingerprint and another two 37 marker McKenzie submissions that would probably test positive as well. If the scope of L643 included 37 marker submissions, the scope of high fingerprint matches would grow from only nine submissions to twenty-two submissions.

Summary of L643 testing

Dominant Surnames (positive): Matheson(1) and Mckenzie (1) and Orem (1).

Year that SNPs were discovered: Apri, 2011.

Source of SNP: FTDNA WTY - Orem, FTDNA ID N1851 and Y-Search ID GZ6XG.

Number of Positive Tests: Three known.

Number of Negative Broad Tests (under 50 % match): 17 found.

Number of Negative Tests within Fingerprint (over 50 % match): None known.

Possible Untested Candidates (over 70 % match): Around six.

Pending Tests (within Fingerprint): None known.

TRENDS OF TESTING CANDIDATES

Note that the number of known testing candidates does not have the typical trend for L21 SNPs where the number of testing candidates declines down to 70 % matches and then sees a sharp rise as non-L643 submissions dominate the under 50 % matches. There is a radical drop from a perfect fingerprint match to very few testing candidate below a perfect match yet above 70 % match. This implies that L643 is more genetically isolated than most SNPs and probably will have less breadth than most SNPs. However, none of the 89 the transitional submissions (between 50 and 70 %) have been tested to date.

ISOGG Status - Prove genetic and surname diversity of L643

L643 satisfies both the 15 % genetic diversity requirement and the surname diversity requirement for inclusion on the ISOGG haplotree. Two submissions of L643 must have at least a genetic distance of 10 (15 % x 67 markers = 10).

DNA FINGERPRINTS

L643 Fingerprint (L21 Off Modal Mutations):

Markers 1 to 37: 389-2 >= 30, 449 <= 29, 607 <= 14, 570 <= 16, and 442 <= 11 (5 mutations).

Markers 38 to 67: 534 >= 16, 444 >= 13, 481 <=21 and 572 <= 10 (4 mutations).

Markers 38 to 67: 572 <= 10 (1 mutations).

Note: The above signature is missing the key marker 481 <= 21 and is much too broad of a signature for L643.

(from MRCA of positive submissions - not WTY):

Orem, N1851, 8 out of 9 match and GD = 6.

Closest Negative Test Known (67 markers)
(from MRCA of positive submissions):

McKnight, 29523, 4 of 9 match and GD = 10.
