Analysis of L1336 SNP

SUMMARY OF SNP - March, 2013

Classification of SNP: Small branch. L1336 is a son of DF21 and has no known sons. L588 and L1337 appear to be equivalent SNPs of L1336. This analysis is speculative in nature since the nature of the curve of the testing candidates suggests major overlap of L1336 submissions with non-L1336 submissions.

Scope of Testing within Fingerprint (67 markers): With over 100 testing candidates, much more exhaustive testing is required to determine how much overlap of positive and negative submissions exists at lower fingerprint matches. The scope of L1336 is probably 20 to 30 submissions but testing will be very mixed at fairly high fingerprint matches. L1336 should qualify for the ISOGG haplotree with minimal testing.

Dominant Surnames (positive): Moore and Gregory.

Source of SNP: FTDNA - WTY testing FTDNA ID 120616, Y-Search ID GPUVF, Surname Moore, discovered November, 2012.

Number of Positive Tests (67 markers): only two known.

Number of Negative Tests below 50 % matches (67 markers): Twelve known.

Number of Negative Tests within Fingerprints (higher than 70 %): One found at 90 % of fingerprint.

Tests between 50 and 70 % matches (67 markers): None tested to date.

Pending Tests (within Fingerprint): Possibly one or two submissions.

TRENDS OF TESTING CANDIDATES

L1336 Fingerprints

The curve associated with the testing candidates does not appear to be typical for L21 SNPs where the number of testing candidates declines down to 70 % matches, remains low between 50 and 70 % and then sees a sharp rise as non-L1336 submissions dominate the under 50 % matches. The number of testing candidates immediately begin to rise as the number of fingerprint matches decline and then flutuates up and down as the fingerprint match decreases. L1336 will probably test positive and negative up to 90 % of the fingerprint but all 100 % matches should test positive. 456 <= 15 appears to be required to test positive for L1336.

ISOGG Status - Prove relationship of L1336 to other L21 SNPs

Requirements of proving L1336 position on the ISOGG haplotree have not been met and L1336 has not been added to the ISOGG haplotree. Below is a summary of SNPs tested:

ISOGG Status - Prove genetic and surname diversity of L1336

L1336 does not satisfy the 15 % genetic diversity requirement but does satisfy the surname diversity requirement for inclusion on the ISOGG haplotree. Two submissions of L1336 must have at least a genetic distance of 10 (15 % x 67 markers = 10). With minimal testing, L1336 should qualify for the genetic diversity requirement.

DNA FINGERPRINT & SIGNATURES

Note: The fingerprint could require fine tuning as additional testing reveals the true scope of L1336.

L1336 Fingerprint (L21 Off Modal Mutations):

Markers 1 to 37: 389-2 >= 30, 464c <= 16, 460 <= 10, H4 <= 10, 456 <= 15, 607 <= 14 and 576 <= 17 (7 mutations).

Markers 38 to 67: 590 >= 9; 413b >= 24 and 534 >= 16 (3 mutations).

Note: 456 <= 15 may be required for testing positive for L1336 and 576 <= 17 is another key marker value that most times will probably required to match as well.

L1336 Signature (R-L21 Plus): 5909-A (closest match)

Markers 1 to 37: 449 <= 28, 464c <= 16, 460 <= 10, H4 <= 10 and 607 <= 14 (5 mutations).

Markers 38 to 67: 590 >= 9 and 617 >= 13 (2 mutations).

This signature will have to be greatly modified to match the L1336 fingerprint.
