Analysis of L1065 SNP

SUMMARY OF SNP - February, 2013

L1065 is also known as CTS11722. Classification of SNP: Large branch. L1065 is a son of L1335. L1065 does not share the second Price fingerprint that L1335 has. L743 is proven to be a son of L1065. L1334 is currently known as a very small branch off of L1335 and probably is a son of L1065 as well (but not proven to date). This analysis is somewhat speculative in nature since the nature of the curve of the testing candidates suggests a significant overlap of L1065 submissions with non-L1065 submissions at lower fingperprint matches. With only twelve known submissions tested out of over 600 good testing candidates, the true scope of L1065 is not known. The only difference between the L1335 fingerprint and the L1065 fingerprint is GATA-H4 >= 12 (not found in L1065). For L1335 positive submissions, 62 % are GATA-H4 >= 12. For L1065 positive submissions, only 33 % are GATA-H4 >= 12. CTS6838 and CTS7030 are currently equivalent to L1335, however, the second Price fingerprint of L1335 has not been tested for CTS6838 and CTS7030.

Speculative preliminary estimated breadth of SNP: 542 = 217 x 95 % (8 of 8) + 256 x 90 % (7 of 8) + 117 x 85 % (6 of 8) + 59 x 10 % (5 of 8).

Scope of Testing within Fingerprint (67 markers): With over 600 testing candidates, much more exhaustive testing is required to determine how much overlap of positive and negative submissions exists at lower fingerprint matches. Analysis supports only one fingerprint and L1065 has tested negative for the second Price L1335 fingerprint which is very small in scope.

Dominant Surnames (positive): Many surnames.

Source of SNP: FTDNA - Believed to be discovered by FTDNA, first tested positive by FTDNA ID N5620, Y-Search ID NSP69, Surname Backstrom, discovered around November, 2012.

Number of Positive Tests (67 markers): Twelve known.

Number of Negative Tests below 50 % matches (67 markers): Sixty-eight known.

Number of Negative Tests within Fingerprints (higher than 70 %): None.

Tests between 50 and 70 % matches (67 markers): Only one positive test known to date (only includes 5 of 8 matches).

Pending Tests (within Fingerprint): Probably many via Nat Geo 2.0 tests.

TRENDS OF TESTING CANDIDATES

L1065 Fingerprint

The number of known testing candidates appears to be typical for L21 SNPs where the number of testing candidates declines down to 70 % matches, remains low between 50 and 70 % and then sees a sharp rise as non-L1065 submissions dominate the under 50 % matches. However, low fingerprint matches have much higher counts than most L21 SNPs which suggest significant overlap of testing positive and testing negative at lower fingerprint matches. L1065 will probably test positive for almost all submissions down to 70 % matches, have a significant mixture of positive and negative test results between 50 to 70 % matches and then test only negative for below 50 % matches. Below is a chart that summarizes the only known fingerprint of L1065:

ISOGG Status - Prove genetic and surname diversity of L1065

L1065 satisfies both the 15 % genetic diversity requirement and the surname diversity requirement for inclusion on the ISOGG haplotree. Two submissions of L1065 must have at least a genetic distance of 10 (15 % x 67 markers = 10).

DNA FINGERPRINT & SIGNATURES

The L1065 fingerprint may require fine tuning as additional testing reveals the true scope of L1065. Currently, there remains only one fingerprint associated with L1065 and broad testing may yet discover more fingerprints.

L1065 Fingerprint (L21 Off Modal Mutations):

Markers 1 to 37: 391 <= 10, 389-2 >= 30, 458 >= 18, YCAIIb >= 24 and 456 <= 15 (5 mutations).

Markers 38 to 67: 531 >= 12; 413a <= 22 and 444 <= 11 (3 mutations).

L1065 Signatures (R-L21 Plus): 1335-SC

Markers 1 to 37: 391 <= 10, 389-2 >= 30, 449 >= 30, H4 = 12 and YCAIIb >= 24/22 (5 mutations).

Markers 38 to 67: 531 >= 12; 413a <= 22 and 444 <= 11 (3 mutations).

L1065 still may a subset of the 1335-SC signature and more testing is required to determine if L1065 could be a duplicate of the primary fingerprint of L1335.
